Microrheology of hyaluronic acid-based 3D scaffold materials for tissue engineering

Tissue engineering requires biocompatible scaffold materials whose open-porous, three-dimensional architecture enables an adequate supply of nutrients to the cells. The mechanical properties of the matrices play a fundamental role not only in differentiation but also in the regulation of cell division and cell metabolism, because differentiated tissue cells such as keratinocytes and fibroblasts can also perceive the hardness and elasticity of their direct environment (mechanosensing) and adapt their metabolism to these conditions. Since in a macroporous, liquid-filled system the local rheological properties do not necessarily correspond to the macroscopically accessible parameters, the Multiple Particle Tracking method is used to determine microrheological properties of the network structure as well as the pore fluid. Thus, the direct environmental conditions in the network and in the pores can be evaluated locally in hyaluronic acid collagen cryogels.
